No. Unlike some software that requires license files to be placed in specific directories, MIDI Guitar 2 does not require you to move the license file into any particular folder. You simply point to it during activation, and the software stores the activation status internally. The original license file can remain wherever you saved it—it does not need to be moved.

Download your .license file from the order confirmation email and save it to a location you can easily remember. The Downloads folder is convenient, but for permanent storage, consider moving it to your Documents folder or another secure location.
